Currently, we are working on an Alzheimer’s disease (AD) imaging project. Briefly, we designed and synthesized some beta-amyloid plaque avid agents. These radioligands were labeled with 99mTc. The in vitro results were quite satisfying. Right now we are trying to perform Autoradiography. We obtained AD and normal brain samples and stained them with Thioflavin. However, the results are somewhat confusing. Based on the pictures how we can determine the beta-amyloid plaques?

two techniques for amyloid one IHC the other tinctoral.. The Gongo red is a classic for amyloid, ensure that you have a known positive/negative controls for any stain... Sorry but based on the pictures, I wouldnt ;-)
Hi Sajjad, I agree with Peter. ThT is good for monitoring fibrillar structures of isolated proteins. Congo Red birefringence is the classical amyloid stain for tissue samples.
