Hello Dear scientists, I am doing Bacteriocin related research, I did a lot of activities to isolates Bacteriocin producing LAB and I used NaOH, catalase( Sodium thiosulfate as catalase) to remove inhibition activities due to organic acid and Hydrogen peroxides, but still couldn't isolate Bacteriocin producing LAB, to get CFS , I used 10,000- 13,000 rpm for 10-30min, with maximum volume of 2ml Eppendorf tube, even to precipitate the Bacteriocin , Ammonium sulfate was used, to extract again different solvents like chlorom, ethanol, methanol were used. but still I couldn't isolate and extract Bacteriocin . So, please would you help me , what are the possible solution, may I follow to extract the potential producers.

What is your target pathogen ?
-It will be more challenging in case of G-ve bacteria.
-Try to use large volume to produce CFS (30 ml) will be helpful for (pH Adjustment and enzyme addition).
-Start screening using the neutrilized CFS only.then go for precipitation with ammonium sulphate& other steps.
