There are several techniques that can be used to identify bacterial species to the genus level, even if you do not have access to an automated DNA sequencer. Here are a few options you might consider:

Biochemical tests: Many bacterial species can be identified based on their unique metabolic properties, such as their ability to produce certain enzymes or their sensitivity to specific antibiotics. There are numerous kits and assays available that allow you to test for these characteristics, which can help you identify the bacteria to the genus level.

Microscopy: Observing the bacteria under a microscope can also provide some clues about their identity. For example, the shape and size of the bacteria, as well as the presence of certain structures such as flagella, can be used to differentiate between different genera.

Molecular techniques: If you do not have access to an automated DNA sequencer, there are still some molecular techniques you can use to identify bacteria to the genus level. For example, you could perform PCR (polymerase chain reaction) using specific primers that are designed to amplify a region of DNA that is conserved within a particular genus, and then run the amplified products on a gel to visualize the DNA bands. Another option is to use DNA-DNA hybridization, in which you probe a sample of DNA with a labeled probe that is specific to a particular genus.

If you are interested in a precise taxonomy classification, 16S rRNA gene is still the efficient approach for that. It is used to reduce the burden charge of NGS. You can use the short-read sequencing platform or even better the longest one.
