I will be propagating MS2 with E. coli. According to ATCC, a #271 broth is recommended for this. This consists of (for one liter): 10g Tryptone, 1g Yeast Extract, 8g NaCl, 15g agar, plus glucose, CaCl2 and thiamine. I was planning to buy the components separately, but it may make more fiscal sense to buy LB broth. The first portion of this is very close to a Lennox LB broth that I have found. However the main difference is that the yeast is in a higher concentration than the 271 medium (5g/L instead of 1g/L).

So, my question is, will this elevated yeast concentration have a measurable effect on E. coli or MS2 growth. I plan to quantify MS2 using double layer agar assay if that has matters at all.
