I currently doing my bachelor thesis at a biotechnology company.

One of my tasks is to establish a method for the quantification of peptid nucleic acids by UV spectroscopy. DNA and RNA are measured by UV spectroskopy at 260nm and 280nm. For a sequence of pnas of known type and number of bases the amount of substance can be determined by using the molar absorption coefficients of the bases at 260nm. However, not all of the nucleobases have their maximum at 260 nm; moreover, the influence of the polyamide backbone on the absorption maximum is unknown. I couldn't found anything for this issue. If someone could provide me a paper dealing with this or similar issue it would help a lot. Thanks a lot!
