Most common practise is by acid hydrolysis (50-64% H2SO4 ; 20-75 C ; 30-240 min), etc....you can obtain NCC with different treatment (ultrasonic, homogenization, acid hydrolysis with HCL or mixture of HCL and H2SO4) and combination of those above, but this is the most common one. For start, try : add MCC in 58-62 % H2SO4 (precooled) 1:10 w/w , temperature: 45 C and time: 60 min. After that, add DI H2O in double weight to stop hydrolysis, then centrifuge it at 4-6000 rpm for 10 min, repeat this for 4-5 times until pH is ~4. You have now solution of NCC...depends of your need , you can further proceed with dialysis or freeze / thawing, or use next method as combined one etc.
To test if you achieve NCC, make 0.5% NCC in DI H2O, and steer it a bit. Let it stay for 24 H, if your solution doesn't get cleared, you succeeded !
