I wish to perform an experiment which will provide support for a novel DNA structure. Most of the experiment is conceptually simple, easy to perform, and inexpensive. But the key to doing it is the counting of plasmid chromosomes. At one time this might have been done by electron microscopy, but very few EM techs know how to look at DNA anymore. I thought later that maybe fluorescence microscopy might be useful, but I cannot find anyone who counts plasmid chromosomes by FM. Now I'm thinking 'particle counter'. There are a number on the market, and they are all unaffordable, by me as least. If you have a particle counter, and think that you can count 4-5 kb plasmid chromosomes, I'd like to talk to you.
