Is there a commerically available plasmid with an ori and selection marker (preferably autotrophic) to express your protein of interest under the Ptet promoter in yeast? So far I can only find the CMV plasmids for integration.
At some point I have been using collection of tetO7 plasmids for yeast published in Gari et al Yeast. 1997 Jul;13(9):837-48.
Introduced gene is expressed under ADH1 promoter - it was strong enough to me.
I do not know about commercial availability but I can help if you experience any problems to get it.

the plasmids indicated by Tomasz are excellent for overexpression, which seems to be your point. However, they have an issue of significant leaky expression of the cloned gene in repressing conditions.
In case zero leakage in repressing conditions is a requirement for your experiments, you may consider the plasmids described by the same group in Belli et al 1998 Nucleic Acids Res. I regularly use pCM252, which has a very tight control of expression. Hope this is useful

Alesandro is right about the pCM series of plasmids. You can get them from Euroscarf for research (http://web.uni-frankfurt.de/fb15/mikro/euroscarf/data/tet_plas.html). I hope that its useful.
Thanks - see all these plasmids are available on EUROSCARF so no problem to get them. A non-leaky plasmid would be best so I'll try the pCM252.
