Is there a commerically available plasmid with an ori and selection marker (preferably autotrophic) to express your protein of interest under the Ptet promoter in yeast? So far I can only find the CMV plasmids for integration.
the plasmids indicated by Tomasz are excellent for overexpression, which seems to be your point. However, they have an issue of significant leaky expression of the cloned gene in repressing conditions.
In case zero leakage in repressing conditions is a requirement for your experiments, you may consider the plasmids described by the same group in Belli et al 1998 Nucleic Acids Res. I regularly use pCM252, which has a very tight control of expression. Hope this is useful
