Dear colleagues, we are trying to purify RNA-polymerase from A. baumannii, but our usual protocol (that works for many other species) doesn't work. Do you have any experience in this field?
Hi, be aware that disrupting A. baumannii cells is very difficult. Maybe what it is not working is the lysate preparation. If you are using a french press, you need to go for a higher pressure than with E. coli and pass the cells three or four times. Use cells from early exponential phase, no more than 0.25 OD600. Soft methods of extraction such as sonication or enzymatic rupture do not work well. I hope this helps.
