I am working on S. aureus ATCC 6538 strain. I prepared electro-competent cells and tried to electroporate the plasmid into this strain. After many attempts, this strain failed to accept the plasmid. I tried to use phages phi80, phi11, phi52A and phi85 to transduce the plasmids. Some plasmids showed transduction when treated with phi85. But majority are not getting transduced properly into this strain. Hence, I am request you to suggest any other alternate methods to transfer the plasmid to this notorious ATCC 6538 strain. Thanks in advance.
Try conjugation here is a reference (although it occurs at a reduced rate from gram -ve to gram +ve it might still solve your problem). Also keep in mind that how your prepare your plasmid (what ecoli strain you use) will have an impact on success due to restriction modification systems.

Staph is not really easy to electroporate you should use strain RN4220, which has been genetically modified to accept foreign DNA. Once you have your plasmid on RN4220 then you can transduce it yo your ATCC6538 strain.
