I am looking for a recipe of alkaline yeast culture medium, and hope the pH could be adjusted to 10~11. Can I just use yest malt agar with pH at 11? I also need to add antibiotics such as Chloramphenicol into the medium to prevent bacteria growing.
Thanks for your question. In fact, I am not quite sure whether ant yeast or mold living in the sample I am studying on whose pH is above 10. I want to prepare some alkaline medium and try to isolate them. Maybe there is no yeast in such high pH surroundings, I want to test it.
OK, so I guess just adding a buffering system to yeast media should be OK (HCO3Na/NaOH for instance)... But i don't think you will isolate any yeast from this media.
