I want to co-culture Neutrophils with leukemia cells (non-adherent) and want to separate the Neutrophils from the leukemia cells after the co-culture. If Neutrophils are purely adherent then I will be able to do that.
The degree of adherence of neutrophils to glass or tissue culture plastic surfaces will depend on the stage of differentiation of neutrophils. If the neutrophils are fully mature, then they will readily adhere to the surface. Neutrophils show a tendency to adhere under serum free conditions to glass or tissue culture plastic surfaces, and especially quicker in response to stimulation. Supplementation of culture media with 10% fetal calf serum or 1% BSA counteracts neutrophil adhesion to surfaces.
