I have heard that there can be a good deal of variability in reported RNA concentration and RIN for repeated measures of the same sample. In one case, the reported concentration was 10-fold different. Has anyone had a similar experience?
I am considering using the RNA 6000 Nano Kit to determine the integrity of RNA I've isolated from various mouse organs and tissues before running the samples on a Fluidigm gene expression chip.
Hi Nick we find the analyser pretty consistent. We did have a problem similar to what you described but it disappeared when the electrodes were cleaned. We actually have two separate electrodes now - one we use for DNA and the other for RNA as DNA measurements can make the electrode quite "dirty" and affect the more sensitive RNA measurements. Also the measurements are more consistent with the Nano6000 chips when the RNA concentration is between 25-50 ng per well (with a max of 100 ng so dilute more concentrated samples).
