I am currently testing the GeneAid DNA extraction kit on abalone (Haliotis iris) attachment muscle tissue samples and it was working pretty fine, until I added the RNase A treatment (as stated in the same protocol) and that's when the DNA concentrations started to be very low. I know that some RNase A are not DNase free, so I boilt the RNase A at 95C for 15 mins first and then I centrifuged it and removed the supernatant so that I could obtain a DNase free-RNase... but I still get very low DNA concentrations. Has anyone had to deal with the same kind of situation?
