I tried BCA with liver extracts, but the most accurate was LECO (BCA could be affected by EDTA and other anti-coagulants
For LECO of blood and liver: homogenization of liver tissue (i used ultraturrax). Then freeze-dry liver homogenate and blood samples to get the powder..Once in powder proceed with LECO machine (put 1 g or 2 in aluminium paper and place it in the machine)...the LECO method you'll just need somebody familiar with your equipment to help you do it the first time.
You do not need to mash/grind. Just take a sample of the liver/blood (usually 0.2 g) and digest it with Conc. Sulphuric acid/catalyst. You could try several samples to ascertain accuracy of the results. Mashing is likely to cause some loss of protein by failure to completely remove sample from the "mashing container". However, you could compare this with the method suggested by Garcia for accuracy.
