I am working on A549 cell line. I am working with my synthesized molecules along with two standards anicancer drug as positive control.
I have few queries related to my results.
1. With doxorubicin i got dose related toxicity from 0.156 to 2.5 uM two fold serial dilution [toxicity is 20%(0.156uM) to 80% (2.5uM)]. but 5, 10 and 20uM shows similar toxicity and which is less toxicity compare to 2.5uM. (Toxicity is 80% for 2.5uM and 60% for 5, 10 and 20uM). What is the possible reason for this trends, i have repeated this experiment and i got similar trends.
2. Docetaxel shows similar toxicity for all concentration range of 0.156uM to 20uM (two fold serial solution). Toxicity is about 35 to 40%.
Both drugs are working fine on CAL-27 cell line. Both the drugs gives IC50 in namomolar and trends have good dose response curve.
Cell seeding are 5000/well in 96 well plates. After 24h of incubation drug treatment was given. Again incubated for 48 h. Alamar blue solution was aaded and incubated for 3h and fluorescence was redaed at 560ex/590em wavelength.
Can anyone give probable reasone or solution for above queries.
