I am looking for CD24 Ab for rat. unfortunately, the most antibodies are available for human and mouse. Does anybody know how to look for antibodies similarity between rat and mouse?
Do the datasheets say something about cross-reactivity with rat antigen? This is the only way to know that for sure. Or testing cross-reactivity by yourself, but you would need to have the antibody and the antigens in your hands.
Even though you can detemine the aa identity % between antigens from different species, cross-reactivity for a particular antibody (monoclonal) is difficult to predict. If you know the exact epitope recognized, you can look for conservation of this fragment in the rat antigen, but since recognition often depends on the structure, this has to be analyzed with caution. If you dont know the epitope, nothing can be predicted.
If the antibodies are polyclonal and there is a high aa identity between the different proteins, there should be some level of cross-reactivity, but the extent also has to be determined experimentally.
The rat IgG2b constant region is most homologous to mouse IgG2a/b and rat IgG1 and IgG2a constant regions are most homologous to mouse IgG1 constant regions, and rat gamma 2c resembles mouse gamma 3. Such a high similarity between rat and mouse immunoglobuin constant regions requires considerations when secondary antibodies are used in double labelling experiments or in cases where mouse antibodies are used in rat tissues or vice versa, since antibodies against a rat or mouse immunoglobuin are likely to cross-react.
