Any tips for gating Hoechst 33342 vs. Pyronin Y fluorescence to identify quiescent cells?

Hi everyone,

I'm seeking advice on the best gating strategy for analyzing Hoechst 33342 vs. Pyronin Y fluorescence in my flow cytometry data. I'm particularly interested in identifying a quiescent population but am unsure how to distinguish between low and high Pyronin Y content in a rigorous manner.

Any tips or recommendations on gating strategies and data analysis would be greatly appreciated! Thank you in advance.
