Whilst testing patient plasma against a panel of HCV pseudoparticles, my negative control which is a patient who has not been infected with HCV is repeatedly neutralising, therefore indicating HCV-specific antibodies. However this control in E1/E2 ELISAs shows no binding. Does anyone have any suggestions why this is occurring and a possible way to stop this happening?
I guess you have PCR amplicon contamination? try using Cobas HCV Amplification and detection kit. The reason to use this is Roche Amplicor system uses Uracil N glycosylase system that degrade PCR amplicon in the samples and then perform selective single step reverse transcription and amplification
Asim Shaghil Thanks for your response but it is not pertaining to PCR amplicon contamination. It is to with non-specific neutralisation of HCV pseudoparticles by the negative control plasma. I have found a work around where I spin the sample down.
