I am trying to histologically slice rat lungs. I am trying to mount either in OCT or paraffin but am looking for something to fix the tissue but also inflate the tissue. Previously, I have used to OCT/Sucrose solution for inflation and then mounted in OCT ... is this the best way? Or is there a way to inflate, fix, and embed in paraffin.
Inflate the fresh lung with 10% buffered formalin from the main bronchus. Then, close the latter to avoid formalin pouring and put the whole specimen in a jar with formalin. After complete fixation, you can process to paraffin as usual.
That's a simple and effective way to fix organs in distension.
