I have tried NucleoSpin® Plasmid and Qiagen Mini-Prep Kit and got just between 30-70 ng/ul DNA from 4 ml culture in both case. The plasmid I am trying to isolate is high copy. Do you think I should increase the amount of culture? From my past experience, I had managed to get 1.3 µg/µl concentration using same protocol I used for high copy plasmid (while working with fungus). This time it didn't work. Expecting critical suggestions.

I believe you shall try to design an OD curve at first to understand the best moment when to use your culture.

I would also suggest you to try a Midi prep to increase your consistency.
