I am having problems fixing the mucus layer on both mouse colon sections and human colon cell line (which secretes mucus) grown on transwell.
I am using methanolic carnoy's fixative, which is recommended in many publications, and have tried fixation times ranging from 10minutes to 3 hours and more.
Methanolic carnoy's fixative has been recommended as the best fixative but my mucus layer simply seems to come off once the fixative touches the sample.
Any advice would be very helpful, thank you so much.

I think you should try acetic acid. I have aleady had this problem and, to my knowledge, this is the only way to reduce the action of the goblet cells...
