Hello guys am trying to lyse Acanthamoeba cells (cysts). As you all know they are very resistant to any type of lysis buffer. Anyone familiar on how i can lyse these cysts without denaturing the contents. am looking at the ATP contents in these cells.

you can try ultrasonic homogenizer , and you can keep the temperature suitable by ice .

this will depend on the instrument you have , you can use its manual to select the proper volume , duration and frequency .
