Can you share your protocol? We pretreat cells 4hrs before transfection with 25mM chloroquine, increases the transfection efficiency many fold!!

Thanks for your concern, this protocal is the one i do without using viral package with few GFP expression: Split the cells one day before transfection in DMEM/10% FBS medium in 6well dish. Prior transfection bring all reagents to room temperature. Add 500ul of DMEM without FBS and PS to 1.5ml EP tube. Then add 7.5ul of 1ug/ ul PEI reagent. incubate the mixture at room temperature for 5minutes. Add the required concentration of plasmid to the EP tube with DMEM and PEI reagent. incubate at room temp for 30minutes. while waiting for incubation, bring the six well plate with grown cells that have reached 60-75%confluency, wash the cells one time with PBS and then add 2ml of DMEM media free from FBS and PS to each hole.After incubation add the mixture of DMEM, PEI reagent and Plasmid dropwise to each well. then swirl the plate and incubate in the cell culture incubator. after six hours replace the media with complete DMEM with FBS and PS. Wait for 48hrs to observe the GFP on microscope and harvesting of the cells.

I will share another protocol about transfection when i use lentviral, but this one i got nothing about GFP expression, i normally use 10mg/ml of Polybrene to facilitate transfection but i get nothing until this moment.I have never used Chloroquine.
