I am planning to use Poly-ornithine/Laminin coating to coat coverslips and grow iPSC-derived neurons. I see there is a big range of Laminin products, some of them quite pricey, and I was wondering if 1) anyone can recommend a specific laminin reference that seems to be consistenly working well and 2) anyone knows whether the presence of mg and ca in balanced salt solutions used for laminin preparation (eg HBSS) affects laminin coating. Hi Maria,
- Laminin (murine, EHS-derived) – Sigma Aldrich #L2020 or Recombinant CellAdhere™ Laminin-521 (STEMCELL Technologies, #77003)
- Prepare laminin in cold buffer (PBS or HBSS); Mg²⁺/Ca²⁺ in the buffer doesn’t harm coating, but keep conditions consistent and avoid repeated freeze-thaw.
- For stable adhesion without frequent re-coating, DendroTEK dPGA | Protease-Resistant Culture Matrix provides a durable base that can be combined with laminin overlays. Use this instead of PDL/PLO and get rid of clumping and peeling!
