You can use placket burman design for optimizing media components by taking enzyme activity (Responsible for reduction) and/or protein concentration (Responsible for stability of nanoparticles) as outcome variable. For parameters, you can select those parameter which will influence the extracellular enzyme activity and help to maximize protein concentration. During synthesis of nanoparticle you can optimize for pH, incubation time, temperature, rpm and metal ion concentration using factorial design for optimization.

If you mean of getting supernatant than you can use high speed cooling centrifuge at the maximum rpm which your centrifuge tubes can withstand.
I can centrifuge with 15000 rpm.after centrifuge we separate supernatant and add 1mM Agno3 to supernatant,after 4-5h color change to brown.your advise for me about my work.thanks
