I am working with a cell population that requires fixation and permeabilization for its identification. At the same time, I would like to assess intracellular ROS/oxidative stress levels. Since I cannot avoid the fixation/permeabilization step, I am looking for a fluorescent probe that can reliably measure ROS in fixed cells.
Most common ROS probes (like DCFH-DA) lose signal after fixation, so I was wondering if anyone has experience with dyes (e.g., CellROX series or other fixation-stable probes) that remain compatible with fixation/permeabilization.
Could anyone suggest a suitable probe or share protocols that worked in similar situations?
