Could you advise me about TEM imaging for studying bacterial-liposomal interactions?
I aim to acquire a comprehensive understanding of how my Liposomes (1) adhere/fuse with bacterial cells (empty formula) and (2) affect the structure/morphology of bacterial cells (antibiotic-loaded liposomes at MIC & Sub-MIC).
I have a few questions:
Is it possible to image the impacted bacterial suspension directly after loading it on a copper-mesh grid without fixation?
Or is it sufficient to load the bacterial-liposomal suspension on a grid -> Fix it with fixation solution -> image it--without embedding the sample in a resin block?
I would appreciate any insights you can offer. Thank you in advance for sharing your knowledge and expertise with me.
