I always prefer to use an inflammatory stimulus that is relevant to how the cell detects the challenge in te first place (e.g bacterial LPS) rather than a generic downstream activator (e.g. TNFa). I worked on inflammation in primary keratinocytes and found the inflammatory response to bacterial antigens triggered more relevant cellular pathways than with just TNFa. Check out Pam₃Cys-Ser-(Lys)₄, Hydrochloride (Sigma 506350) which can stimulate IL-8 from both proliferating & differentiated keratinocytes.

Mark Ian Fowler. Thank you, I have just noticed your comment! I couldn't be sure that LPS will induce the interest cytokines, so I ordered TNFa, which seems the best stimulus for ILs (based on the LRs). hope it works effectively.

I will update the comment when I use it.
