Hello, my Ap antibody (A2951, Sigma) can be used on fixed frozen crosections, but I can not get the positive resutls on paraffin sections after trying many times. I need some suggestions, especially from those who used this antibody on paraffin sections before. Thanks a lot.

Does the datasheet say anything about the use of the antibody on FFPE material? If an antibody works on frozen tissue, does not mean it will work on FFPE sections.

I did a quick search for you on BenchSci for A2951. I found a paper that used this antibody in frozen sections.

I'd suggest searching through BenchSci for additional antibodies that have been published in frozen section IHC.

I have not used this particular antibody, but it may be worth trying the antigen retrieval step. You can place the slides in a coplin jar filled with sodium citrate buffer (10mM sodium citrate, 0.05%Tween 20, pH 6) into an oster steamer for 45min, then allow to cool for 25min and wash with PBS. This should be done after blocking and prior to exposing to the primary antibody.
