Hi everyone,

I want to design a protocol to detect viral RNA of Hepatitis C virus though real time PCR with probes. I used to use a regular qPCR with primers against the 5'UTR region (most conserved among hepatitis subtypes) and the dye SYBR, but I would like to try a probe to improve the sensitivity and specificity of the detection. There is a kit (COBAS® AmpliPrep/COBAS® TaqMan® HCV Test) commonly used for diagnosis, but I would like to have my own probe and primers in the lab. Obviously, they don't give the sequences of the primer&probe from the kit.

So, anyone has worked with some designed primers&probe against HCV (1b or ideally all subtypes) that could recommend me to use? there is some publications indicating probes, but I would like to know of some near experience.
