I bought an amyloid beta 1-42 and it has 95% purity. I usually dissolve 1mg in 1 ml 100% TFA (vortexed and sonicated),after evaporation by nitrogen I resuspend it in 10 mM HFIP and evaporated again and repeat that 3 times to ensure we have monomers.
The important point now is the peptide dose not dissolve in TFA from the first step and I can obviously see fine particles suspended in it.
The supplier said it should be soluble because they used TFA in synthesis process .
So what should I do?
If it has formed fibrils already it can be insoluble in acid. You can try HFIP or HFIP together with some alkali.

I know you probably don't want to modify your procedure right now and specially working with amyloid. But since that particular batch is giving you some trouble maybe you could try dissolving it forming the ammonia salt using 1% solution of ammonium hydroxide in water. We do it this way when we run our AB 1-40 experiments but some other groups also do it with AB1-42.

Also, which is the company you are buying the amyloid from?
