I'm working with INS-1 cells that have been co-treated with a cytokine cocktail and different plant extracts for 24 hours, followed by harvesting the cells to determine protein expression by means of western blots. The cytokine cocktail has been optimized in INS-1 cells to induce a moderate state of inflammation as inflammation is associated with amylin expression.

After probing for amylin, my bands appeared at about 50-75 kDa; as indicated on the attached image. Previous studies have reported amylin expression at a much lower MW (+/- 10kDa), while other studies that looked at amyloid expression got bands showing up at a range between 85 - 130 kDa. Does anyone perhaps have a suggestion as to why the bands appear at a much higher MW when I probe for amylin and not amyloid?
