Hi, due to a mistake of a person in charge of receiving the deliveries, I have only just realised that Agencourt RNAClean XP beads for the purification of cDNA (RNAseq application) were kept at -20 even though the packaging says DO NOT FREEZE and Keep at 4 degrees.
Do you know how freezing would affect the performance of the beads? Would it be still alright to use them? They were in -20 for about 4 months now.
Freezing will mess up the bead matrix in a similar way that freezing an agarose gel messes up its matrix so that you can get DNA out of the agarose gel by the Freeze and Squeeze process. I would not use the beads for anything important.
I think you should try using it for single sample and check the results. As it contains oligo dT beads so just first thaw it in room temperature without any vortexing for about an hour. When it liquefies and attains the room temperature then you vortex it for about 15 to 20 minutes before use to get a homogeneous mixture of Ampure beads.
I agree with Romit. I would suggest to increase the volume by 5% to increase clean up chances.
