yes but be careful about over amplification.remember 10 cycles of pcr is 1000 times more product so use very little target and few cycles. I would use 1ul of a 1;1000 dilution of the first amplification product and set up tubes of pcr and after about 8 cycles remove 1 tube every 3 cycles up to about 20 cycles and see what works. After too many cycles the produc anneals to itself and you will just get a smear of long product not a band. A better way is to use nested pcr where you make new primers just inside your old ones and use these to re amplify a clean product. for instance if your primers are 1-20 and 200-180 of a 200 base product then nested primers could be 10-30 and 190 -170. the second primers do not have to be perfect primers as the target sequence is very small compared with the original genome
