This is strictly dependent on the expression vector you are using to transform your target DNA in E. coli DH5 alpha. For example, if you are using Blue/White screening method, then you can see blue colonies (transformed with native intact bluescript vector) and white colonies (transformed with the modified vector encoding your target gene). Other colonies without vector will die as you add an antibiotic (for example Erythromycin; Erm) to the LB media.
