Using two different antibodies we observe an accumulation of LC3B-I not LC3B-II when cells are treated with Ivermectin, and this is exacerbated by Bafilomycin A1 addition. We compared FCCP which gives LC3B-II appearance? .
Also probed for Parkin which drops expression going from control extracts to IVM+BafA1 treated extracts. Any explanation for LCB3-I accumulation?

Rather than LC3-I accumulation, I think you should think that the expression (transcriptional) level of LC3 increased.
Given that LC3 conversion was not observed, you should check the expression change in p62/SQSTM1 instead to determine whether autophagy is activated or not.
