We are looking to knock down a transcription factor in kupffer cells, preferably with AAV to allow cell type specific cre-dependent miRNA expression in lysMCre mice. Thus, we would appreciate shared experiences with AAV transduction of Kupffer cells or other macrophages. Which serotypes should work, if any?

I am not an expert and maybe my answer will be not the best one. I am using AAV for retina infection and what I can tell you is that usually for almost all the cell types the REP genes from AAV 2 are the best used. For the serotypes then it is really according with your cell type, because normally is the CAP gene to give the tropism.
In this optic I am using a very good serotype called as AAV 2/ DJ where the REP gene comes from AAV 2 and CAP gene is coding a chimera of 8 different cap proteins coming from different AAV types. This system is improving in different tissues and organs a lot the yield of infection.
You could check this one because they should have also a list of different cell types infected with the yield of infection.
