I am now planning to deplete the NKT cells in c57 mice and then injected with tumor cells. I searched online, and found some people used NK1.1, some used CD1d. I am now confusing as NK1.1 antibody also deplete NK cells. Can anybody give me a protocol or share some experience for NKT cell depletion, such as antibody, dose, time for good depletion and time for tumor cell injection? Thanks a lot.
Hi, NKT cell depletion is tricky indeed, especially if you need to spare NK cells from depletion. NK1.1, unfortunately, is neither specific nor sensitive, i.e. the NK1.1-negative (i)NKT cells will be left. There is one report on a depleting antibody directed against the invariant TCR of NKT cells (Scheuplein, ... Schaub. PLoS One 2015). That may work depending on your experiment.
