I have a dataset pertaining to weight of female verses males from different populations. I wish to do a two-way ANOVA to find variation at sex and different population level. Also i want to know how to enter data in the matrix ?

There are loads of tutorials in Youtube especially on 2-way ANOVA. Since you have two independent variables, you will need to do further analysis (post hoc). If the interaction is significant, then it would be better if you could do separate analysis for each separate group. It can be a little confusing at first, but start by going through some videos first.
