Hey, all

I intend to do XPS depth profiling to find the localization of the drug inside albumin nanoparticles. The synthesized NP-encapsulated drug is roughly 200 nm in diameter. What is the ideal method of sample preparation for this?

While there are number of XPS studies on protein/albumin nanoparticles coated on surface of gold or any other metal alloy available in literature (Article XPS study of protein adsorption onto nanocrystalline alumino...

) there seem to none where pure albumin or albumin nanoparticles were used.

I intend to drop cast the sample on the surface of a sterile glass slide and send it for analysis. Is this the correct method? Will the presence of sulphur (sulphur being highly volatile) damage the instrument? How is XPS analysis of sulfur-containing compounds usually done?

Thank you all In advance

Best
