I am not sure if you are using the color-coupled assay for TAGs quantification.

In my PhD work, I did check the TAGs of male and female Drosophila, which varies a lot based on different genetic background, age, and reproduction status. Normally virgin female flies have higher fat content than males.

40-60ug/fly (female), 10 (or less) -30 ug/ fly (male) are the TAG level obtained in my hand. If you have interests in the CCA method, you might check the paper (https://www.nature.com/articles/s41598-019-43327-y)

thank you for your answer. I indeed find similar levels with the color-coupled assay.

However I was using this paper for reference: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5752231/

Where they seem to find 100µg/mg female fly, which is more or less double (female flies weigh about 1 mg). I suppose the method of homogenization of the flies might play an important role in how much fat is released?
