I transformated my sequence confirmed insert positive clone in pET6xHN vector into BL21DE3 cells, however I accidentally spread the transformation on LB/amp plates that also had XGal and IPTG. Will having the XGal and/or IPTG affect downstream protein expression?
Xgal and IPTG are transient activators of the lacZ system so they won't affect your cells unless they have that gene construct. Once you find colonies, simply streak them out on a new plate without Xgal and IPTG and you will be just fine.
