I am growing Penicillium chrysogenum and Penicillium brevicompactum in a liquid batch of potato dextrose and streptomycin. I am trying to make them sporulate. By increasing the incubating temperature (20 degree C to 25), increasing exposure to light and placing them in a shaking incubator, will I see changes in sporulation in the next few hours?

Hi Morgan: If you target is to harvest spore, I recommended you to grow the fungus on PDA, not broth, Also to enhance sporulation light period is important. Also but in your mind, the more rich medium gave less sporulation and more biomass
Yes I am trying to make the fungi sporulate and I am having a hard time. I see lots of mycelia growth but no spores. I have plated them on 25% PDA plates, I am hoping to see spores after a few days. If you have any other advice to accomplish sporulation I would appreciate it! Thanks!

Penicillium sp. sporulate at the air/liquid(or solid) interface, so the usual procedure is to grow them on agar plate medium of some type. If you really need to use liquid medium for some reason then you don't want to shake it at all - the mycelia will form a mat and sporulate from there.
I haven't grown the two species you are working with, but if you still have trouble getting them to sporulate on the PDA I would try adding a trace element and vitamin solution to the agar - sometimes sporulation requires elements that aren't needed for mycelial growth.

Hi: You will get sporulation after 3-4 days from the inoculation on PDA,
