I am working with high molecular weight (HMW) Hyaluronic acid (HA) (more than 2MDa) and I functionalized it with tyramine (HMW HA-TA) . Degree of substitution is 5%.
I measured the viscosity of HMW HA and HMW HA-TA at 1.3-1.5% concentrations and the decrease in all is too much. I used PBS as the solvent and after vortexing, I allowed it mix at 4deg for 48hrs.
What could be the possible problems? How can I solve the problems please?

Dear Alma Banigo, more possibly thé tyramine actes as a spacer side group, reducing mutual intermolecules cohesion favoring thus good solvency. This of cource when molecular degradation during modification is excluded. Tg measurements in this case give better insights. My Regards
