Recently i have been trying to measure colour difference between my samples. I am a little confused as to why i cannot just use the basic colours - i.e., red, green and blue to measure the colour difference between the samples. Why is there a need to use the formulae from International Commission on Illumination (CIE) for the measurement instead? I am not really sure how to explain the need to have "brightness or luminosity" as a variable for the measurement? How do i explain this?

It all relates to what you want to find. The values of R, G, and B do not give a definite answer to whether one or another color is closer to your target color. Can you be sure that R=0.6, G=0.7, B=0.3 is green? Or is it actually yellow? Or going further, is R=0.3, G=0.7, B=0.6 better green than the one before?
That's why you use color difference based on CIELAB color space, where you can specify your target color and have a direct answer on the color difference. In CIELAB you can also set any color, not just primary colors as it is in RGB color space.
If you are not satisfied with CIELAB and you just want to have a simple comparison, you can also use 'color quality', which is based on hue difference, value, and saturation of the color, and compares your obtained color to primary RGB/CMY colors.
