I am working on A431 cells and since few days the cell pellet after trypsinization and centrifugation is becoming purple instead of white. The cells in culture are looking healthy and are without contamination. Is it common to get a colored cell pellet? Kindly help me in this regard.

What I feel is that your culture media might have some issues. The components of culture media are adhering to the cell surface or reacting the membrane moieties and thereby probably the cell lines appearing purple instead of white. I think you must use freshly prepared media to culture your fresh experimental cell line.

You might be trypsinizing it longer, for which surface proteins getting denatured and capturing the dye from the media. If your cells are growing in mono layer, they should get up within 1 mins of trypsinization followed by little tapping. Try trypsinizing for lesser time and see if the problem can be fixed.
