I have isolated phosphate solubilizing bacteria (PSB) from soil using total plate count method with Pikovskayas Agar Media and incubated for 72 h at 30 degree Celsius. They have showed good Phosphate Solubilization Index (PSI) and robust growth. The obtained bacteria was cultivated in Nutrient Agar slant for 72 h at 30 degree Celsius and preserved at 4 degree Celsius for working culture. However, the PSB didn't show any growth in Luria Broth medium after I cultivated two loops from the slant. Can someone explain? Should the working culture preserved only in 30 degree Celsius?
Note: I have preserved the PSB culture in NA slants for almost a week.

@ Ghaida, I think you can store the soil at 4 degree Celsius but after isolation of PSB, the temperature should be maintained between 28-30 degree Celsius even for the working culture to grow.
I think at 4 degree Celsius the growth of the living entity restricted, except archaebacterial species most of the eubacterial performance high at 25 to 35 degree Celsius.
at 4 degree Celsius most of the metabolic activity inhabited and the growth of the bacteria restricted.

I do not understand the answers to this question. So far as I can see, the questioner has isolated the bacteria at 30C and then stored it on agar slants at 4C, which is usually reasonable. Amrit Kumar's answer appears to assume that the subsequent cultivation of the stored slant is also at 4C, which is not reasonable (except for eg Listeria). Of course growth and metabolism are restricted at 4C but they should return to optimum at 30C. I do not know this PSB bacterium but unless it is known to be susceptible to low temperatures (unusual) then there must be another reason for its failure to regenerate at 30C, possibly desiccation (the osmolarity of the medium might be too high too). Sometimes it takes a few days for stored bacteria to "wake up" so perhaps incubate at 30C for longer?
