I believe it is because a change in a specific protein is not inheritable, as the information is actually in the DNA (in the case of cells). If the 'inheritance of acquired characteristics' hypothesis was right, any change in that protein would result in that same change in the offspring - which does not happen, since that new mutated protein information never goes back to DNA or RNA.
