I am performing EvaGreen dPCR assays on the QIAcuity One with known concentrations of template DNA to confirm the assay's dynamic range, but the resultant concentrations are significantly lower than the expected (as low as 16x below). In previous runs with these same known concentrations, I got higher results (still lower than expected), or else I would be more dubious of my resuspensions and dilutions.

Would anyone be able to shed some light on what might be affecting my results?

Thanks in advance!
